Okay, I saw two of the most famously improving replacements:
1) Reba McEntyre as Annie Oakley in AGYG. She was all they said she was and lit up the stage. Brent Barrett was a wonderful replacement Frank Butler. He was a wonderful replacement in everything I saw him in including "Chicago" and others.

2) Heather Headley as Shug Avery in the revival of "The Color Purple" - though I am sure Jennifer Hudson sang it well.

3) The original Broadway production and the Signature revival of "Angels in America" and "Perestroika" by Tony Kushner had some remarkable replacements. In the original: Cynthia Nixon as Harper Pitt, Jay Goede as Joe Pitt, Dan Futterman as Louis, Cherry Jones as the Angel/Nurse et al. and F. Murray Abraham as Roy Cohn. Signature revival: above all the sensational Michael Urie as Prior Walter, Keira Keeley as Harper Pitt, Jonathan Hadary as Roy Cohn and a sexy Adam Driver as Louis. That replacement cast was remarkable.
