A couple of weeks after Kim Stanley stopped showing up for the 1964 production of Three Sisters, Geraldine Page moved from playing Olga to taking over Stanley's role as Masha.

Certainly actors who have understudied a role and played a smaller role in a production have moved to the lead role they understudied. For example, three Tevyes during the original run of Fiddler had started as Avram and understudy for Tevye.

Also during the original run of Fiddler, Mimi Turque started as a replacement Hodel and then came back as Tzeitel.

On the first national tour of My Fair Lady, Michael Evans started as Zoltan Karpathy and understudy for Higgins. When Brian Aherne left, he took over as Higgins and played it for a long time (and then for a while on Broadway while Michael Allinson was sick).

I have a friend who was in the first national tour of You're a Good Man, Charlie Brown. He started as the replacement male understudy, then took over as Linus, and then took over as Snoopy.

In the first London production of Company, Marti Stevens was originally Sarah and understudy for Joanne. She had also done that on the national tour. When Stritch left the London production after six months (as required by British Equity), Stevens took over as Joanne.

In the original production of Sweet Charity, Helen Gallagher was Nickie and understudy for Charity. After playing plenty of performances as Charity, she took over as Charity around June 22, 1967. On June 30, it was made official that she had taken over and it was announced that the show would run with Gallagher through the rest of the summer, but it closed two weeks later.

Rip Torn started as Tom Junior in Sweet Bird of Youth and took over as Chance Wayne for the last month of the run and then played Chance on the tour.

In the original production of 1776, people switched around a good deal. David Ford started as Hancock and then took over as Dickinson. Dal Richards was originally Josiah Barllett and understudy for Rutledge. Near the end of the run, he took over as Rutledge. Edmund Lyndeck started as Witherspoon in the original cast and later took over as Dickinson on tour. There was lots of other moving around among the cast, although ibdb is incorrect that David Cryer not only played Rutledge for a year but also played Jefferson at some point. That wrong info comes from Theatre World, which simply made a mistake. I wrote them about this years ago and they never changed it because they wanted proof. It can be very difficult to prove that something didn’t happen.
